Imaging mass spectrometry (IMS) has developed into a powerful tool allowing label-free detection of numerous biomolecules in situ. In contrast to shotgun proteomics, proteins/peptides can be detected directly from biological tissues and correlated to its morphology leading to a gain of crucial clinical information. However, direct identification of the detected molecules is currently challenging for MALDI–IMS, thereby compelling researchers to use complementary techniques and resource intensive experimental setups. Despite these strategies, sufficient information could not be extracted because of lack of an optimum data combination strategy/software. Here, we introduce a new open-source software ImShot that aims at identifying peptides obtained in MALDI–IMS. This is achieved by combining information from IMS and shotgun proteomics (LC–MS) measurements of serial sections of the same tissue. The software takes advantage of a two-group comparison to determine the search space of IMS masses after deisotoping the corresponding spectra. Ambiguity in annotations of IMS peptides is eliminated by introduction of a novel scoring system that identifies the most likely parent protein of a detected peptide in the corresponding IMS dataset. Thanks to its modular structure, the software can also handle LC–MS data separately and display interactive enrichment plots and enriched Gene Ontology terms or cellular pathways. The software has been built as a desktop application with a conveniently designed graphic user interface to provide users with a seamless experience in data analysis. ImShot can run on all the three major desktop operating systems and is freely available under Massachusetts Institute of Technology license.  相似文献

14-3-3 proteins are highly conserved in species ranging from yeast to mammals and regulate numerous signalling pathways via direct interactions with proteins carrying phosphorylated 14-3-3–binding motifs. Recent studies have shown that 14-3-3 proteins can also play a role in viral infections. This review summarizes the biological functions of 14-3-3 proteins in protein trafficking, cell-cycle control, apoptosis, autophagy and other cell signal transduction pathways, as well as the associated mechanisms. 大型煤电基地(CEBs)是我国煤基能源和材料集中开发区域和国家能源战略安全重要支撑,合理评价CEBs开发生态影响是CEBs可持续协调开发、科学管理中的难点问题。研究将CEBs开发视为系统工程,与自然生态系统(NES)相关联,深入研究大型煤电基地生态系统(CEBES)的驱动行为、内在结构及主要关系、生态影响途径、生态累积过程和生态累积效应及阈值分析方法。基于CEBs空间和功能定位提出CEBs的概念、内涵和特点,融合构建的CEBES涵盖到自然资源、能源开发行为(煤、电等)、其他人类活动(农、牧、城)和大气、水、土壤及生物域;研究表明CEBES的生态累积状态受三种行为驱动和影响(自然行为、能源开发行为和其他人类活动),通过融合、传导和辐射耦合作用,表现为生态要素损伤、系统结构变化和系统状态失衡三级状态,生态累积具有影响多态性、空间多尺度和过程渐变性特点;明确了生态累积过程、累积效应和阈值及其相互关系,引入反映生态影响累积程度的生态损伤系数,提出四类生态阈值(生态要素、内部结构、系统状态和行为调控);基于系统性和实用性筛选反映生态累积相对状态的60项指标,构建具有“三力”驱动、分区要素耦合和系...  相似文献

The conversion of the Brazilian savannas for pastures and agricultural use has caused the species Dimorphandra wilsonii (Fabaceae-Caesalpinioideae) to become isolated and restricted to areas occupied by African grasses of Urochloa sp. This highly endangered tree species was cultivated in the presence of Bradyhizobium japonicum (symbiont 1), Glomus etunicatum (symbiont 2), and Urochloa decumbens in low nitrogen (N) availability in order to evaluate its growth in these experimental conditions. Even though the nodulation and mycorrhization was of low occurrence, the inoculated plants with symbionts had the greatest nitrogen and chlorophyll content, photosynthetic radiation use efficiency, and biomass accumulation in relation to the plants which had not been inoculated and/or cultivated in the presence of U. decumbens. The results suggest effective N2 fixation, independent of the localization of bacteria, whether in the root tissue interior or free in the rhizosphere. Therefore, the presence of N2-fixing bacteria can benefit the early growth of D. wilsonii, whereas the occurrence and aggressive persistence of U. decumbens can limit this development, increasing the threat of extinction of this species in their habitat.  相似文献

The evidence for the existence of receptor heteromers opens up a new field for a better understanding of neural transmission. Based on our theory, we have discovered main triplets of amino acid residues in cell-adhesion receptors of marine sponges, which appear also as homologies in several dopamine D2 receptor heteromers of human brain. The obtained results probably mean a general molecular mechanism for receptor–receptor interactions in heteromers originated from the lowest animals (marine sponges).  相似文献
